Principle of the Test
Virus isolation is the most common method used for the routine diagnosis of Enterovirus infections. A pan-enterovirus antibody directed against a conserved VP1 epitope common to Enteroviruses has been produced. This offers the potential to use a single reagent to detect the presence of an Enterovirus isolate in cell culture monolayers.
The IMAGEN Enterovirus test reagent contains a single pan-enterovirus monoclonal antibody conjugated to a fluorescein derivative. The reagent is used in a one-step direct immunofluorescence technique. Cells inoculated with patient samples are fixed with acetone then stained using the IMAGEN Enterovirus reagent for 15 minutes at 37°C. Excess reagent is washed away with phosphate buffered saline. The stained areas are then mounted and viewed microscopically using epifluorescent illumination. If an Enterovirus is present then characteristic apple green intracellular fluorescence is seen within infected cells, which contrasts with the red counterstaining of uninfected cells.
