Description
Listeria monocytogenes  infection is frequently a-symptomatic.  However, in certain individuals – the very young, the very old and mothers-to-be, for example – it can cause a life- threatening disease known as listeriosis.  L. monocytogenes is typically found in the environment. It can also grow at low temperatures and so can survive refrigeration.  Because of these traits and the serious nature of the disease, many manufacturers test for this organism in their food products. International guidelines have been set up which require manufacturers to show due diligence in ensuring that their product is free from Listeria monocytogenes and often demand absence of the organism when testing samples.

L. monocytogenes in the environment is often present in mixed populations of other Listeria species.  Therefore, many manufactures prefer to look for the genus Listeria in their production environment and/or product rather than just  L. monocytogenes. The presence of any Listeria species is a strong indication that L. monocytogenes could be present.

When Listeria is present in a food environmental sample, the genetic-based BAX System uses the power of the polymerase chain reaction (PCR) to rapidly create (amplify) millions of copies of a unique, Listeria -specific DNA fragment for reliable detection. BAX System assays simplify the method by combining all the PCR reagents you need into a single tablet, conveniently packaged inside the PCR tubes contained in each kit. Reducing the number of manual manipulations and pipetting steps, the tabletted reagents improve accuracy and consistency; hands-on time is reduced to a minimum and shelf life is increased.

After standard enrichment, samples are lysed to break open the cell walls and release DNA. This lysate is then used to hydrate the PCR tablets. Processing in the automated thermo-cycler takes less than 4 hours for a full rack of 96 tests. Electronic results appear as positive/negative icons on-screen, and the data can be easily stored, shared or printed.

Applications
The BAX system detects Listeria on a variety of surfaces, including ceramic, plastic, wood, iron, concrete and more. BAX system Listeria medium (CM1067) combines a single enrichment step with shorter incubation times (24 hours for swabs and 28 hours for sponges) for quicker time to result. Validation studies have shown that the single-enrichment BAX System performs as well at 24-28 hours as the dual-enrichment USDA-FSIS culture method at 48 hours. With specificity rate of >99%, the BAX system for detecting Listeria has been certified by AOAC Research Institute as a Performance Tested method.
