Cefpodoxime Combination Disc Kit

For the detection of ESBLs (extended spectrum β-lactamases) in Enterobacteriaceae which don’t produce inducible AmpC enzymes.

Description
Extended spectrum ß-lactamases (ESBL’s) are found in many Enterobacteriaceae and arise by mutations in common plasmid-mediated ß-lactamases (especially TEM and SHV), which give an increased antibiotic resistance. Oxoid Cefpodoxime Combination Disc Kit contains discs impregnated with cefpodoxime and the ß-lactamase inhibitor, clavulanic acid.

The kit contains the following:

  • Cefpodoxime/clavulanic acid (CD01) 10/1µg
  • Cefpodoxime (CPD10) 10µg

Technique
The discs need to be placed on Mueller-Hinton (CM0337) with sufficient space between the discs to allow the formation of clearly defined zones of inhibition. The use of other susceptibility test media needs to be validated by the customer.

Use an inoculum level 0.5 McFarland suspension on Mueller-Hinton Agar. The plates are incubated at 35-37°C for approximately 18 hours in ambient air.

An organism is interpreted as producing an ESBL if there is an increase in zone size of ≥5mm between the combination disc compared to that of the cephalosporin alone. For example:

Limitations of Use
The CD01 combination discs are designed for the in vitro detection of ESBLs in Klebsiella spp, E. coli and other organisms not producing inducible AmpC enzymes.

Quality Control
To ensure the compatibility of the two components, it is recommended that quality control is performed with the following control strains.
